Intestinal Alkaline Phosphatase Combined with Voluntary Physical Activity Alleviates Experimental Colitis in Obese Mice. Involvement of Oxidative Stress, Myokines, Adipokines and Proinflammatory Biomarkers

Intestinal alkaline phosphatase (IAP) is an essential mucosal defense factor involved in the process of maintenance gut homeostasis. We determined effect moderate exercise (voluntary wheel running) with or without treatment IAP on course experimental murine 2,4,6-trinitrobenzenesulfonic acid (TNBS) colitis by assessing disease activity index (DAI), colonic blood flow (CBF), plasma myokine irisin levels and adipose tissue expression proinflammatory cytokines, markers oxidative stress (SOD2, GPx) adipokines mice fed a standard diet (SD) high-fat (HFD). Macroscopic microscopic sedentary SD was accompanied significant decrease CBF, increase tumor necrosis factor-alpha (TNF-?), IL-6, IL-1? leptin mRNAs mRNA adiponectin. These effects were aggravated HFD but reduced exercising animals, potentiated concomitant IAP, especially obese mice. Exercising demonstrated substantial for adiponectin intestinal mucosa mesenteric fat as compared to animals. The SOD2 GPx significantly decreased mice, these reversed administration. Our study shows first time that combination voluntary oral synergistically favored healing inflammation, strengthened antioxidant ameliorated colitis; thus, may represent novel adjuvant therapy alleviate inflammatory bowel (IBD) humans.
